Cultivation methods and precautions of activated sludge bacteria

Cultivation of activated sludge There are many methods, but different methods need different training time, manpower and material resources. The cultivation method should be selected according to the wastewater quality, climatic conditions and actual conditions.

1. Preparation before training
(1) Construction of each structure and removal of construction waste through clear tanks. The static pressure test proved that there was no leakage and no sinking displacement. Finally, the relevant regulations were passed.
(2) All electrical equipment, machinery, pipelines, etc. are completed and tested through single machine test and linkage test. Finally, it complies with the relevant regulations (instructions).
(3) According to the needs of future operation and management, conditional sewage treatment plants (stations) need to conduct basic routine laboratory tests, such as pH, water temperature, COD, do and biology, to guide the process and daily work of cultivating activated sludge. Run.
(4) Survey basic data, including diurnal variation of sewage flow, water quality (pH, water temperature, COD, BOD5 / CODCr, nitrogen, phosphorus, toxic substances, etc.) and their changes, technical and equipment parameters of various facilities. It is best to record the background water quality of the receiving water body, such as the river receiving the sewage, so as to provide the basis for the impact on the receiving water body after several years..
(5) According to water quality conditions, necessary nutrients (carbon source, nitrogen source and phosphorus source) should be prepared to prepare for deficiencies. The inoculation method also needs to prepare dry (or concentrated) sludge with similar sewage properties as other sewage treatment plants (stations) as the strain for microbial culture of activated sludge.
(6) Operators should be familiar with the pipeline layout and practical aspects of the whole system to understand the basic process and control requirements of sludge cultivation.
(7) After the personnel are in place, the system should be operated continuously after planting and domestication, and can not be removed.
(8) Prepare the necessary original records of test and operation as well as the initial establishment of rules. From the beginning of training, we will gradually establish a more standardized organization and management mode to ensure the orderly start-up and formal operation.
 
Natural culture
Natural culture of bacteria is also known as direct culture. This is a kind of cultivation process that uses a small amount of microorganisms in wastewater to propagate gradually. For this kind of culture method, we can consider the urban sewage and some of the nutrients with low toxicity industrial waste water Such as edible plants and meat processing plant wastewater, but the culture time is longer. Natural culture bacteria can be divided into two types: intermittent culture and continuous culture.
(1) Intermittent culture. Add wastewater into the aeration tank and expose it to boredom (i.e. only aerate without entering the wastewater). After a few days, stop aeration and stand for 1 hour. Then discharge about 1 / 5 of the upper wastewater and inject the same amount of fresh sewage. In this way, the three processes of suffocation, static sinking and water inflow were repeated, but the water inflow increased from the previous time, and the suffocation time was shorter than that of the last time. In spring and autumn, sludge can initially be planted in about two to three weeks. When the sludge concentration of the aeration tank mixture reaches about 1g / L, continuous water inflow and aeration can be carried out. Due to the low concentration of sludge in the initial stage of cultivation, the sludge accumulated in the sedimentation tank was also less, and the reflux was less. Then, with the increase of sludge amount, the amount of returned sludge also increased. When the sludge concentration reaches the concentration required by the process, the normal operation can be started and controlled according to the process requirements.
(2) Continuous culture. First of all, the aeration tank is full of waste water, and then the water is stopped. The water can be poured continuously from half a day to one day. After continuous aeration, the amount of water increases gradually from small to large. After continuous operation for a period of time (similar to batch method), activated sludge will appear and increase gradually. When the sludge quantity in the aeration tank reaches the concentration required by the process, it shall be controlled according to the process requirements.
As the natural culture method is directly using wastewater to cultivate activated sludge, so the cultivation process is the same. Microorganisms gradually adapt to the nature of wastewater and obtain domestication process.
 
3. Inoculating strains
Inoculation culture method is a common activated sludge culture method, which is suitable for most of the activated sludge Industrial waste Water treatment plant. This method can also be used to shorten the incubation time if there is mud near the municipal sewage treatment plant. There are two common inoculation methods:
(1) Concentrated sludge inoculated with bacteria. The concentrated sludge from a nearby sewage treatment plant is used as a cultivated strain (seed sludge or seed sludge). Activated sludge culture and complete nutrition of urban sewage, low toxicity industrial wastewater treatment system can be directly added to the wastewater to be treated for aeration until the sludge turns brown. At this time, the sedimentation tank is also put into operation to make the sludge circulate in the system. In order to speed up the culture process, unfermented dung water or other nutrients can be added in the culture process. When the activated sludge concentration reaches the process requirements, the cultivation process is completed. Economically, the amount of seed mud should be as small as possible. Under normal conditions, the concentration of diluted mixed sludge should be controlled at 0.5g/l or higher.

3. Strain inoculation

(2) Dry sludge inoculation. "Dry sludge" usually refers to the sludge cake which is dehydrated by dehydrator and has a moisture content of about 70-80%. This method is suitable for remote areas, and the transportation distance of sludge is far.
The process of inoculating dry sludge with sludge is basically the same as that of thickening sludge. In order to inoculate the sludge, fresh cake which has just been dehydrated should be used first. Add a small amount of water, then pour it into the mud, and then add it to the aeration tank. The dosage of dry sludge is usually 2 to 5% of the tank volume.
Dry sludge may contain a certain concentration of chemical agents (for sludge conditioning). If the dosage of the drug is too high and the toxicity is high, it is not suitable to use the seed mud as culture. In order to determine whether a small amount of sludge can be put into a small container (e.g. a small amount of water can be added to a container) and a small amount of water can be added to determine whether a small amount of sludge can be put into a beaker. After a period of inoculation, the color paste can be used.

Precautions for sludge cultivation:

(1) The pH, COD, ammonia nitrogen, dissolved oxygen in aeration tank, sludge settling performance and other indexes in influent water should be measured in the process of cultivating activated sludge. After the initial formation of activated sludge, the biofacies were observed, and the sludge culture state was evaluated according to the observation results, and the cultivation process was dynamically adjusted.
(2) The culture of activated sludge should be carried out in the season with suitable temperature as far as possible. Because of the suitable temperature, the microorganism grows fast and the culture time is short. If the cultivation can only be carried out in winter, the inoculation method should be adopted, and the sludge required should be larger than that in spring and autumn.
(3) In the cultivation process, especially after the initial formation of sludge, attention should be paid to prevent excessive automatic oxidation of sludge, especially in summer. This has happened in many factories. This not only increases the time and cost of bacteria culture, but also may lead to the sewage treatment system can not be put into operation on schedule. In order to avoid the self oxidation of sludge, controlling aeration and aeration time is the key. In order to measure the dissolved oxygen content in the water tank frequently, it is necessary to enter the water in time to meet the nutrient requirements of microorganisms. If the fecal concentration is too low to add nutrients if necessary. Intermittent aeration conditions can be used if not available.
(4) In the late stage of activated sludge culture, proper discharge of some aged sludge is conducive to the further growth and reproduction of microorganisms.
(5) The industrial wastewater treatment plant is in production. There is usually no wastewater entering the plant before it is put into production. Once the production plant is put into production, the discharged wastewater needs to be treated in time. At this time, the sterilization time should be reasonably determined according to the actual situation, and the sludge and nutrients should be prepared in advance.
(6) If the sludge in the aeration tank has matured, but there is still no wastewater entering, the aeration should be stopped to make the sludge dormant, or intermittent aeration (extending the aeration interval and reducing the aeration volume) should be conducted to minimize the sludge oxidation rate. If conditions permit, the content of nutrients such as feces and non-toxic organic waste (such as canteen) must be increased.
(7) Most sewage treatment plants have two aeration tanks. In this case, the activated sludge can be cultured in the aeration tank and then transported to another adjacent aeration tank for multi-stage expansion. This method is suitable for large sewage treatment plants.
